An initiative to build the theater building was taken by Antonio Manoel de Vilhena who was a Grand Master of the knights. Grand Master Antonio Manoel de Vilhena had outlaid a hefty 2,184 scudi to built the stupendous theater building.

The first theater was enacted on 9th January in 1732 by the artists of the Manoel Theater house. It was a classic tragedy named as 'Merope'. It was penned by Scipione Maffie. All the knights had taken part in the theater. The arena was designed by Francois Mandolin, the chief of knights. The auditorium was semi-circular and horse-shoe shaped when it was built, but later it had undergone several retrieval and renovation works.

Various tragic, comedy melodramas were enacted on the stage by the knights of St. John, Frenches and Italians. Some popular playwright of Johann Adolf Hasse, Nicolo and Baldassare Galuppi were very popular in the early stages in 1792.

The theater was taken over by the Maltese government in 1956 and since then it had undergone many developmental works. It reopened in 1960.
